Mutation Scanning/Detection
Mutation Discovery Using Hi-Res Melting™
(Registration Required)
Hi-Res Melting curve analysis is a rapid, high-throughput, closed-tube method for mutation scanning and genotyping.PCR-amplified products are subjected to a high-precision denaturation process in the presence of the double-strand DNA binding dye LCGreen® Plus on the Idaho Technology LightScanner® Instrument. Nucleic acid melting is tracked by monitoring the fluorescence of the mixture across a defined temperature range, generating melting profiles that can be used to identify the presence of sequence variation within the amplicon.

iPLEX Gold Assay for SNP Genotyping
Vendor-submitted protocol. Sponsored by Sequenom Published in BioTechniques Protocol Guide 2008
The iPLEX assay for SNP Genotyping is the leading application to follow up on Genome Wide association studies and perform targeted genotyping. Recently, a new protocol has been developed on the basis of the iPLEX assay to improve the multiplexing capabilities of the assay up to a level of 40-plexing.
