Despite advances in next-generation sequencing technology, Sanger sequencing remains the ‘gold standard’ for sequence validation. Sanger sequencing is particularly useful for detecting and confirming mutations within the SARS-CoV-2 genome and conducting variant surveillance. This Application Note provides a protocol for sequencing a region of the SARS-CoV-2 genome from nasal swab samples, using a new terminator sequencing dye with analysis by capillary electrophoresis.
